Research

Type 2 diabetes mellitus affects more than 25 million people in the United States and this costs a total of $245 billion in health care expenditure. Chronically elevated plasma glucose levels have been shown to cause insulin resistance in vitro. In the present study I will examine the detrimental effect of hyperglycemia on insulin sensitivity and mitochondrial function in humans by: (1) chronically elevating the plasma glucose concentration in normal glucose tolerant individuals and (ii) chronically lowering the plasma glucose concentration in type 2 diabetic patients with an inhibitor of the renal SGLT2 transporter. The euglycemic insulin clamp with muscle biopsy will be used to quantitate insulin sensitivity and mitochondria function, and the molecular mechanisms responsible for the muscle insulin resistance/impaired mitochondrial function.
